РОЛЬ ПЕРОКСИРЕДОКСИНОВ В РАДИОРЕЗИСТЕНТНОСТИ РАКОВЫХ КЛЕТОК

РЕФЕРАТ

Пероксиредоксины (Prdx) представляют собой семейство тиол-зависимых пероксидаз, участвующих в контроле уровня внутриклеточных АФК, редокс-сигнализации, фолдинге белков и регуляции многочисленных сигнальных путей. В опухолевых клетках Prdx поддерживают редокс-баланс и онкогенные процессы, способствуя пролиферации, выживанию и адаптации к окислительному стрессу (ОС). При лучевой терапии Prdx выступают ключевыми медиаторами радиорезистентности. Вклад Prdx в радиорезистентность обусловлен их участием в восстановлении широкого спектра гидропероксидов, репарации повреждений ДНК, перепрограммировании метаболизма, поддержании фенотипической пластичности, ремоделировании опухолевого микроокружения и регуляции различных форм клеточной гибели (апоптоз, ферроптоз, некроптоз). 

В данном обзоре суммированы современные представления о структурно-функциональных особенностях и регуляции различных изоформ Prdx, а также о механизмах их участия в ответе на действие ионизирующего излучения. Рассматриваются перспективные стратегии преодоления радиорезистентности, включая применение ингибиторов Prdx, их комбинации с таргетными препаратами, а также подходы, направленные на редокс-модуляцию и подавление стресс-адаптивных механизмов опухолевых клеток.
